At a glance
- Semax stimulates BDNF and NGF expression in the brain (Dolotov et al.)
- Selank produced anxiolytic effects comparable to benzodiazepines without sedation
- Both Semax and Selank are dosed intranasally due to short half-lives
- Pinealon is the neuro-bioregulator in the Calm and Clarity stack
- N-acetyl variants are more potent per microgram than the plain peptides
The nootropic peptides worth your time are the ones with a mechanism you can actually name. Two of them raise the growth factors your brain uses to build and maintain connections. A third is a bioregulator. Everything else in this category is mostly hope with a Russian abstract attached.
Semax: the BDNF driver
Semax is a heptapeptide derived from a fragment of ACTH(4-10), and its defining trait is neurotrophic. It stimulates the expression of brain-derived neurotrophic factor (BDNF) across multiple regions of the brain in vivo (Dolotov et al., 2003), and it rapidly and specifically activates both BDNF and NGF gene expression after a single intranasal dose (Shadrina et al., 2007). BDNF is the molecule most associated with learning, memory, and neuroplasticity, so a compound that reliably raises it is a legitimate research tool rather than a stimulant. The Semax dosage chart covers the intranasal protocol and the more potent N-acetyl variant.
Selank: calm without the fog
Selank is a synthetic analog of the immunopeptide tuftsin, and it is the anxiolytic of the pair. In studies it produced an anxiolytic effect comparable to classical benzodiazepines by modulating the GABAergic system (Kolomin et al., 2017), but without the sedation, tolerance, and dependence that make benzodiazepines a poor daily tool. That combination, real anxiety reduction with a clear head, is the whole appeal. The Selank dosage chart has the dosing, and the Selank research page covers the mechanism.
Pinealon: the bioregulator
Pinealon is a short tripeptide bioregulator studied by Khavinson and colleagues for neuroprotective and neuroregulatory activity in research models. It is dosed in short courses rather than continuously, and it is the third leg of a well-known cognitive protocol. See the Pinealon dosage chart and the Semax research page for background.
How they combine: Calm and Clarity
The reason these three keep appearing together is that they cover different jobs. Semax pushes plasticity and drive, Selank takes the edge off, and Pinealon adds the bioregulator layer. That exact logic is the basis of the Calm and Clarity cognitive stack, which is worth reading if you want the full protocol rather than the components.
| Peptide | Primary effect | Route | Evidence |
|---|---|---|---|
| Semax | BDNF and NGF, focus | Intranasal | Multiple rodent and clinical studies |
| Selank | Anxiolytic, GABAergic | Intranasal | Comparable to benzodiazepines |
| Pinealon | Neuroprotective | Subcutaneous or intranasal | Single research lineage |
Tip: Start with one. Semax and Selank feel different, and stacking them from day one makes it impossible to tell which one is doing what. Add the second after a week if the first is not enough.
Purity and sourcing
Nootropic peptides are dosed by the microgram, so identity and concentration matter more than for larger compounds. Verify the certificate of analysis on our peptide lab test database before you buy. Semax, Selank, and Pinealon are available as research vials from Ascension Peptides with 50% off using code ENHANCED, and Semax and Selank also come as ready-to-use nasal sprays from Limitless Biotech with code ENHANCED for sprays. For a vendor we have vetted in depth, see the Ascension Peptides review.
